There are nearly twenty ways to measure SOD activity, generally they are divided into two categories. One category is measures directly, measure substrate consumption rate or product production rate in catalyzing SOD, such as the EPR, pulse radio solution, potassium superoxide, etc. Due to equipment or reagents, such ways are Hard to apply in laboratory. Another category is measures indirectly, the principle is a system produced oxygen, oxygen rise to another reaction easy to detection, quantitative SOD activity indirectly by measure SODs inhibition to this reaction. These ways are in common use. Such as Pyrogallol autoxidation. Such ways have a common drawback, it is curve relationship between enzyme activity and the inhibition rate. This leads to the following two questions.

In order not to increase the workload too much, and produce less errors, enzyme must reach 48-52% inhibition rate.
2. Linearization of Curve For reaching 48-52% inhibition rate, it requires a heavy workload. Solution way is measured 5-7 gradient of enzyme inhibition rate, use mathematical to convert it to a straight line, after that, each sample can only measure once.
